The basic principles of FISH pretreatment and IHC pretreatment are similar. However, because FISH is used to detect chromosomes within cells, the requirements for pretreatment are higher than those for IHC. Kanlume’s fully automated FISH pretreatment system and sample release agent are specifically designed for rapid FISH probes, with dedicated development in the processing procedures and reagent formulations to make them suitable for rapid FISH hybridization. Standard IHC pretreatment does not meet these requirements.

The length of Kanlume’s rapid FISH probes is 300–500 bp, whereas conventional probes have a length of over 600 bp.

Kanlume rapid FISH probes need to be mixed before use. Take them out of the -20°C freezer 10 minutes prior to use. Do not use a vortex mixer. Gently invert the tube by hand 3–5 times, then centrifuge briefly for 10 seconds. The probes are then ready to use.

The assay can be performed, but it will take longer and the workflow is less convenient. Deparaffinization can be done in an oven (preheat for 1 hour at 70°C), while permeabilization and enzyme digestion are performed in a water bath (change water to cool down after permeabilization). This method yields good pretreatment results.

The KL-200 is a fully automated FISH testing system. It requires no manual intervention from unstained tissue slides to the completion of FISH staining, including all FISH steps: pretreatment, hybridization, and post-treatment.
